Ligands of the GABAA Receptor Benzodiazepine Binding Site

چکیده
Benzodiazepine (BZ) binding site ligands are important central nervous system (CNS) drugs. Their numbers and our knowledge of how they interact with the BZ-binding site of GABAA receptors are both rapidly expanding. The GABAA receptor is a member of the ligand-gated ion channel superfamily. In general, it consists of an assembly of transmembrane pentamers of different subunit compositions (1). To date, at least 15 types of subunits (α1–6, β1–3, γ1–3, δ, ε, and π) have been identified using molecular cloning techniques that combine in various combinations to give rise to the different GABAA-receptor subtypes (1–3). The central BZ-binding site is a part of the GABAA-receptor-associated chloride ion channel. Most functional subtypes of the GABAA receptor contain α, β, and γ subunits, with the different receptor subtypes displaying marked variations in their binding capacity to different BZ-binding site ligands (4,5). When GABA, the major inhibitory transmitter in the CNS, binds to a GABAA receptor, the chloride ion flux through the channel is increased. This leads to membrane hyperpolarization that results in a reduction in the excitability potential of the neuron (6). As a consequence, GABAA receptors are the molecular targets of a variety of pharmacologically and clinically important drugs, such as the anxiolytic, anticonvulsant, sedative-hypnotic benzodiazepines, some anxiogenic, convulsant β-carbolines, and the convulsants bicuculline or picrotoxinin. Furthermore, multiple recognition sites that exist within the three-dimensional structure of the various GABAA-receptor subtypes possess the capacity to interact with a host of different ligands (6). These sites include: the GABA site; the benzodiazepine site; the barbiturate site; the general anesthetic site; the picrotoxinin site; and the storage site for GABA. Optimum Conditions of Radioligand Receptor Binding Assay of Ligands of Benzodiazepine Receptors
To obtain drugs which are more selective at benzodiazepine (BZD) receptors, design and synthesis of functionally selective ligands for BZD receptors is the current strategy of our pharmaceutical chemistry department. The affinity of newly synthesized ligands is assessed by radioligand receptor binding assays. Based on our previous studies, 2-phenyl-5-oxo-7-methyl-1,3,4-oxadiazolo[a,2,3]-pyrimid...

متن کامل3-Arylisothiazoloquinols as potent ligands for the benzodiazepine site of GABAA receptors
3-Arylisothiazolo[5,4-b]quinolin-4(9H)-ones and 3-arylisoxazolo[5,4-b]quinolin-4(9H)-ones were synthesized and assayed for affinity for the benzodiazepine binding site of the GABAA receptors. While the 3-arylisothiazoloquinolin-4-ones were found to be potent ligands, with affinities (expressed as the affinity Ki value) down to 1 nM, the 3-arylisoxazoloquinolin-4ones are less potent. This is sug...
متن کاملHypothermic activity of acetaminophen; Involvement of GABAA receptor, theoretical and experimental studies
Objective(s):The mechanism of hypothermia action of acetaminophen (APAP) remains unclear even 125 years after its synthesis. Acetaminophen produces hypothermia. The mechanism of this reduction in core body temperature is not clear but evidence shows that it is not dependent on opioid and cannabinoid receptors. Because of strong documents about the roles of GABA and benzodiazepine receptors in h...
متن کاملAn Updated Unified Pharmacophore Model of the Benzodiazepine Binding Site on -Aminobutyric Acida Receptors: Correlation with Comparative Models
A successful unified pharmacophore/receptor model which has guided the synthesis of subtype selective compounds is reviewed in light of recent developments both in ligand synthesis and structural studies of the binding site itself. The evaluation of experimental data in combination with a comparative model of the 1 2 2 GABAA receptor leads to an orientation of the pharmacophore model within the...
